Biochemical risk protection For protecting the biochemistry risk, please following the below precautions:

Risk of biological contamination Incorrect use of the sample may lead to infection. Do not touch the sample, mixtures and liquid waste directly use hand, please wear gloves, work clothes to avoid being infected when operation, if necessary, please wear protective glasses. If the sample contact with the skin accidentally ,please processing following the user standard immediately. Some agents have a strong acid or alkali. Please be careful using , to prevent the hand and close thing to contact the reagents directly .if the reagents contact with the hands and clothing accidentally, please washing with soap and water immediately. If the reagent gets in the eyes accidentally ,please washing with water immediately and consult an ophthalmologist immediately. 5

Waste Fluid treatment In order to prevent the waste fluid cause the environmental pollution and physical harm, please following below items when dealing with waste fluid.

Risk of biological contamination Some substances from Reagents, Liquid quality control, Calibration solution , Strengthen the cleaning fluid and waste fluid are control by the pollution regulations and emission standards. Please following the local emission standards and consultation with the relevant reagent manufacturers or distributors.

Attention Items In order to operate the equipment correctly and effectively, please pay more attention to the following items. System Usage

Warning This equipment applies to serum, plasma, urine and cerebrospinal fluid. Out of this scope, please consult SUNOSTIK. Please reference other clinical symptom or other test results, when estimate the clinical result.

Environment

Attention Please set the equipment in the appointed environment followed by instruction. Out of the appointed environment, the test result will be incorrect and broke the system. If want to change the system, please connect SUNOSTIK service hotline or local distributor.

Electromagnetic wave and Interference Prevention Attention Please do not put a noise equipment near to the system. Please turn off any electromagnetic equipments in the room, where the system in. Such as mobile phone, radio equipment and so on. And do not use any CRT monitor near the system. Noise, electromagnetic wave can be cause the malfunction and affect the result.

System Use Attention （1）Please performance the equipment followed by instruction. Incorrect use will cause the wrong results; even the system and person will be harmed. （2）First time, please check the control to confirm the equipment is run. （3）After turn off the main power 10 seconds, open again. Otherwise, the system will be protected. If it is, please turn off the main power then turn on again.

System Maintenance Please maintain the system regularly followed by below notes. Otherwise, lifetime and characteristics will be affected.

Attention （1）Please maintain the system followed “System Maintenance” in the instruction. Incorrect maintenance will cause the wrong results, affect its characteristics and lifetime, even system and person can be harmed. （2）The surface will accumulate dust in long time. Use soft cloth to put in the water and twist it, then rub the surface lightly. Immerge the cloth in to soap liquid if necessary. Do not immerge the cloth in ethanol or related solvents. At last, use the dry cloth to clean.Before cleaning, please close the power and pull out the plug. During cleaning, please prevent water goes into the system, otherwise, system and person can be harmed. （3）Do performance the control analysis, after change the main parts, such as source lamp.

Chapter 1 instrument installations Warning SBA-733 Plus are required to install by SUNOSTIK’s engineer or by the people who was authorized by Sunostik company.

1.1 Unpacking On receiving this instrument, check whether the packing is in perfect condition and that the seals are intact. Open the box and take out its content with care (Figure- 1.1). Read this chapter carefully before installing the instrument. Do not throw the packing material away. In case of the re-shipment, use it again to guarantee good transport.

1.3 Temperature and Humidity requirement Temperature should be between 0℃to 40℃(Fluctuations less than ±2℃/H) The humidity for system store should be 30% RH ~ 80% RH and also non-condensing。 If the environment is out of this range, please using air condition to cooling.

Warning If the system is idle for a long time do not use, this may speed up the system’s mechanical and circuit aging process, so run the instrument on a regular time is required.

1.4 Power supply requirement ◆ ◆ ◆ ◆ ◆

230V～, 50Hz, 3-core power cable, good grounding； Well-grounded power outlet to provide the necessary power supply。 Should be has at least 3 meters distance between the instrument and the power supply plug。 The current of power supply plug should be more than 1 A Choose the right fuse

1.5 Waste liquid box  Choose the right length of the waste pipe that you need.  Put one end into the waste liquid bottle.  Put anther end into the waste liquid exit of the equipment. You can see below picture for details.

Risk of biological contamination Make sure wear gloves when install the pipe of waste liquid bottle. Put on work clothes to prevent be contaminated. Put on the protection glassed if necessary

Attention The environment should be ventilation to make the emitting system work normally, if necessary use the ventilation device. But please prevent the air be blew to the system directly, otherwise the data will be affected.

2.3 Membrane keyboard

Three parts of the keyboard: numeric, letter and function keys. Numeric keys: The keys 【0】～【9】and【.】constitute the numeric keyboard. They serve for inputting and revising the numerical data. Letter keys: The keys 【A】～【Z】and【-】constitute the letter keyboard. They serve for inputting and revising the letter parameter information. Function keys: Perform routine work and activate some functions. ◆ 【Clear】to delete the mistype data. ◆ 【Wash】to washing the flow cell, in order to reduce the cross-contamination. The peristaltic pump will be high speed rolling when this key is pressed, at the same time plenty of distilled water will be sipped in to rinse the flow cell, again press the key to stop the operation. This function is suggested to be proceeded before and after the test to reduce the cross-contamination, and improve the test accuracy. ◆ 【】、【∧】、【∨】to move the bar to change the focus. ◆ 【Esc】to exit the program or back to the main menu. ◆ 【Enter】to write or confirm the information. ◆ 【Absorb】the key is in the left front of the equipment, to run the peristaltic pump to sip the sample. ◆ 【F1】、 【F2】、 【F3】、 【F4】match with the words which are displayed at the bottom of LCD separately, different functions in different displays. Blank means no function.

2.5 Thermal Printer

Characteristics:  384dots/line  Resolution 8dots/mm  Max print speed 75mm/s  Built-in thermistor to check the needle of the printer.  Built-in sylvatron to check the paper.

2.6 RS232 Interface Go though the RS-232 interface to connect the equipment with computer. You can program a better report by using the enclosed CD software.

2.7 Pipette and Imbibition Switch When absorb the sample, put the pipette into the bottom of cuvette to prevent absorbing the bubble during do this action. After pressing the switch, the imbibition pump will self-start to absorb the sample into the flow cell. Take away the pipette from the cuvette quickly, when the imbibition device stopped. At this time in order to improve the test accurate, the air will be absorbed in this device, to reduce the cross-contamination between two contiguous samples.

Biological Pollutant Risk When doing the test, please mark sure the gloves and working clothes are wore, if necessary take on the prevention glasses.

Attention When checking the sample, please mark sure the pipette is straight to prevent the result accuracy is affected.

2.8 Colorimetry System Colorimetric system includes peristaltic pump, quartz flow cell (32ul), optional system and mechanism gearing, wavelength adjustment system and Peltier . The system is controlled by CPU, temperature control and wavelength lookup both be proceeded under the CPU. To be Noted that you’d better take out the silicon tube from the peristaltic pump when the instrument will not be used in long time to avoid the tube block.

Warning The colorimetric system is the main part of this equipment, unprofessional untrained person don’t open it randomly. Otherwise the properties will be affected.

High Temperature Warning When change the lamp, mark sure the power is off over than 20 minutes, otherwise your hands must be burned by the high temperature of the lamp.

Chapter 3 Software Introduction 3.1 Self-test After open the power switch, (see ① in the picture of

the equipment back form

The self-test will be started 5 seconds later, to test the memory, printer, filter and sampling circuit separately and display the status real time. The temperature adjustment will be started after the self-test, refer to below picture. Main menu will be indicated after the temperature adjustment, or you can enter the main menu by pressing any key directly. At this time, the temperature adjustment is kept going without any display.

Attention You’d better to start the test after complete the temperature adjustment to make the test accuracy.

Attentions for Err or Alarm: "err: #02 Cannot find flash"：Flash is broken or connection is malfunction. "Warning: #01 Printer has no paper"：No paper in the printer or connection is malfunction. Works can not be affected by this alarm. "Warning: #02 Sample test failed"：A/D read failure. Malfunction of pick-board or the connection is malfunction between pick-board and main-board. "Warning: #03 Open lamp failure"：Lamp is off or low A/D.

3.2 Window Description Window Format: All windows include in this system with the same format, such as main menu(below picture): There are three parts in the window: title bar up the window to display the subjects and time, middle is the customer area to indicate the parameter list and submenu information, status bar down the window to indicate shortcut key and cue information.

Sample Test Main Menu Description: There are 6 submenus under the main menu; cursor is rest on the “sample test” position default. You can move the cursor with keys【】 【∧】 【∨】, the related function also will be indicated, at the same time the related functions will be displayed on status bar. Press【Enter】 to enter the submenu.

Attention Press the shortcut keys【1】~【6】to enter the submenu directly.(From left to right; from up to down). Shortcut key “R” means to delete the results; “P” means to print; “T” means set time; “M” means to A/D; “U” means to update the window; “H” means the basic information.

The following is the brief introduction for each submenu: ⑴Sample Test: It is the beginning of testing one sample. After programming one item, press the key to test. ⑵ Absorbance: It is specially ready for testing the absorbance. You can test the absorbance after enter the menu. The wavelength, temperature, absorption volume and delay time can be programmed. There will be a hint when you exit the submenu. ⑶ Programming: You can write all the programs of 100 tests under this menu. You can also revise the program under this menu. ⑷ Parameter setting: There are four parameters can be set under this menu, such as measure device, sipping volume adjustment, air sipping and language. ⑸ Application menu: There are 6 functions for using, such as patient record, printer setting, time, system maintenance, software update and help. 20

SBA-733Plus Manual ⑹ QC query: Under this menu, you can check the saved QC information for 100 test items. There are 3 QC type under each item, each QC type can save the QC result 100 days. There are 2 modes for QC indication (QC data list and QC chart), both can be printed out. You can evaluate the accuracy of the test items through the QC chart, the instrument also can automatically calculate out the QC data’s average, differentiation and variance of the certain month.

3.3 Parameters setting Under the main menu to select parameters setting or directly press the shortcut key 【4】to enter this window, see as below:

Sample Test Measure device setting The instrument can available for flow cell and colorimetric cup, the default device is flow cell. The samples sipping and draining is controlled by the peristaltic pump under the flow cell selection, convenience and efficiency; the peristaltic pump will stop operation under the colorimetric cup selection, the pump push button should be pressed for confirmation under the manually sample adding. The advantages of colorimetric cup are highly-transmittance and few cross-contaminations, etc.

Absorption volume calibration If there is incorrect, you can adjust the parameter to keep the accuracy. This parameter is direct ratio with the absorption volume, default 100%.

Air sipping setting This parameter is used for setting the length of air, the default is 80uL. It can be adjusted base on the pipette length, air can not be sipped into the flow cell is the condition. 21

Attention The absorption volume calibration and air sipping setting should be correct, incorrect setting will cause the wrong results.

Language The operation language is available for many languages. After selection, it works immediately.

3.4 Application Menu To select the application menu or directly press the shortcut key【5】to enter this window under the main menu, see below picture:

Check the record Application menu instruction There are 6 submenus under the main menu. You can move the cursor with the directional keys by pressing the buttons to select different submenus. At the same time the related functions will be displayed on status bar. Press【Enter】 to enter the submenu. The following is the brief introduction for each submenu: ⑴ Patient record: Enter this window to find the details of the patient test result and test time, to print or delete. See below picture:

Comprehensive print: Input two code numbers and press key 【Enter】to print all the records between this two codes. Lab Name: The default is SUNOSTIK, it can be adjusted according to the customer request, and the name will be printed in the test report. (It is workable with the standard print type). Auto-print: The test result can be auto-printed out after complete the test if this function is in application. Print Style: There are two print type is optional, default is standard type, there is 23

You can select economy type to save the paper, but the result content will be compactness.。 Print Status: The item is for read only to indicate the printer status. Self-test: The shortcut key【F1】is used for testing the paper and checking the printer. Paper Moving: The shortcut key【F2】is used for moving paper, this function can be acted when the printer is installed. Repress it to stop the moving. ⑶ Time System: Using the keys【】to move the cursor to revise the date and time after enter this submenu. See below picture:

When the test results is generally low or too high, you can use this parameter to amend the results.

Attention Default number is 1，To setting this coefficient should be according to the huge statistic data, it should be applied in serious.

4.3

Calibration parameter

Calibration Mode: Edit with the form of sub-window,there are two modes for selection:STANDARD and FACTOR. If you select standard ,the standard liquid should be used during the test. The equipment will calculate the factor and saved it for the future automatically. If you select factor, you can apply the factor for the test directly to save the standard liquid, there is no need to set the calibration. If the test mode is Kinetics, the calibration will be inefficacy and hided automatically. Factor Value: If the calibration mode is Factor, The factor value must be input for calculation. If the calibration mode is standard, the factor value will be inefficacy and hided automatically. Standard quantity: If the test method is multi-standard, that need to set the quantity of the standard liquid, other test method , this parameter will be inefficacy and hided automatically.

Standard Thickness:

If the calibration mode is standard, the standard thickness should be input for calculation. If the calibration is Factor, this parameter will inefficacy and hided automatically. If the test method is Kinetics, this parameter will be inefficacy and hided automatically. The standard thickness will be show X when the test method is multi-standard. 2.

Standard value X:

The standard value X should be input under the multi-standard test method（each thickness of the standard liquid），The maximum range is 2-6 , it is according to the set of standard quantity. Under the non multi-standard method, this parameter will be inefficacy and hided automatically.

Attention The setting of the standard value has to be followed from small value to big value , or it will cause the mistake result.

QC data

Absorb the water The above picture is in case of the measure device is flow cell, and the information indicated in the status bar is “Absorb the distilled water”，please prepare the distilled water and press the Absorbance button, the peristaltic pump start to working. The indicated content will be “absorbing…”. After absorb the distilled water, the status bar will display “withdraw the cuvette ” with one beep. Please take out the pipette from the cuvette at this time, and the analyzer will absorb in the certain air (the air absorbance item is included in the system parameter) to separate the different liquid, In order to avoid the cross-contamination. If the measure device select the “Colorimetric cup”, The above status bar will show “add the distilled water ”.You should put the colorimetric cup full filled with the distilled water and press the button. There is no absorb procedure when use colorimetric cup the mentioned as above. It is the same for adding the sample, we will not mention again. The delay time will be come out after finish the absorbance , in order to level off the distilled water,, and the status bar will show “in delay 00X”,

If CV%（coefficient of variation）exceed 5%，it means

the set-up time of the

analyzer is too short and the analyzer is not in steady, please wait a moment and again try. Or it is caused by the aging of the peristaltic pump，the liquid is in fluid status, please replace the peristaltic pump and try again. 2.

If Mean（Average value）under 150，maybe caused by the bad connection of the flow cell or the colorimetric cup，or there are some air bladder in the distilled water, please deal with it and again try.

If read the baseline successful, it will enter into the different interface base on the selected test item; the following will introduce of the procedure for each test method.

Chapter 7 Maintenance 7.1 Replace printing paper When installing printing paper, please cut the end of paper to a correct shape according to following picture. Extend the printing paper to feeder of the printer.

7.2 Replace fuse Fuse ① is on the rear of instrument ,it was installed together with power button, Rating current is 6A.

1 When replacing fuse, firstly twist off the cap ,remove old fuse and install a new one equipped together with instrument.

Warning If you replaced a new fuse and turned on power ,the fuse was burned at Once .Under this situation ,please don’t change a higher current fuse any More ,turn off the instrument and get in touch with our maintenance engineer at once.

7.3 Replace lamp The instrument uses a 6V/10W halogen lamp ,life span is about 2000 hours ,because the lamp is in high usage frequency , after a period of usage the lamp will be not bright enough or can’t work any more ,at such time ,you need to replace a new lamp. First, twist off the cap, loosen two inner hexangular set screws（2）be used to fix component parts of lamp base （4） with an inner hexagon spanner，remove the component parts and remove halogen lamp （1）from lamp base（3）.Install a new halogen lamp on the lamp base ,and install the component parts on the lamp holder ,fasten the screws with inner hexagon spanner .Please see the following pictures to check detailed installing position.

High temperature warning When replacing lamp ,you must wait 20 minutes after turning off the power ,or your hands may be scalded by high temperature comes from the surface of lamp.

SBA-733Plus Manual Ajust light source： When you need to replace lamp, loosen fixed screws under the instrument to open upper case .Please pay more attentions when removing upper case to prevent electric shock .To place upper case down from the right side(facing the instrument direction). Loosen hexangular set screws with inner hexagon spanner .Install the new lamp as original way .Before fixing lamp base ,assure the strongest light of lamp is just in the centre position .You must assure filament is perpendicular to bottom board .After proper adjustments ,please fix lamp base with inner hexagon spanner.

Adjustment and detection: After fixing lamp ,please monitor A/D Value under maintenance window when standard filter is 340nm, the A/D value should be larger than 120nm at least ,if this value is too small ,please readjust position of lamp .please see below operating interface:

7.4 Replace pump pipe  Peristaltic pump located in the rear of instrument ,If you find the instrument don’t aspirate liquid or the volume disagree with pre-set value(software setting didn’t work),please remove peristaltic pump and check the reason .One possible reason is the pump pipe blocks or a long time using and wearing happens to peristaltic pump self ,both of these make an increasing interspace between shaft and three pulleys ,at such situation ,you need to replace peristaltic pump or pump pipe.  The way to remove peristaltic pump: first ,disconnect the pump from pipeline ,then press the pipe joint outside pump with your left hand ,at the same time ,pull the pump pipe out with hard strength. 43

7.5 Daily Maintenance  Never use cleaning agent or abrasive cleansers, use clean water or fabric dipped with neutral soap .  In case of any reagent or corrosive substances were splashed on instrument, clean it at once with damp cloth and clean water.  It is necessary to clean liquid circuit on correct way after finishing one group of tests and daily job .Clean with plenty of distilled water after finishing one group of tests.  Clean liquid circuit with equipped cleaning agent everyday after work,clean tube with plenty of distilled water circular to empty liquid circuit.  If the end of suction tube is broken ,you could cut down several millimeters vertically and clean the cut part.  If the tube is seriously run of date ,please change a new one and use equipped instrument components  If long time don’t use the instrument ,please empty liquid circuit, then pull out waster liquid tube and store it suitable.  Clean surface of instrument with non-abrasive cleansers every month.  Wipe off dust fell outside the fan every month.  Clean the tube and flow cell after finished tests everyday to avoid dirty precipitate.  Don’t pull flow cell out of instrument unless cleaning the flow cell .Do pull and plug the bolt slightly .also pay attentions to placing position of flow cell ,never install it on oppositely direction and lead to damage.  A canonical operation way could extend service life of instrument .Never press soft keyboards with sharp nail on which way the keyboard could be easily broken .You should press it with finger belly .  When installing printing paper ,if a bit of paper was left in printer ,please don’t pull it out forcibly，because such way could easily damage the printer .The right way is press【Paper】key to send out paper.

Warning Most maintenance were carried on under power off state ,so please maintain it after turning off power for safety.Never spray any liquid like reagent to instrument or electronic accessories.

Biology contamination danger Do wear gloves and cloth to prevent from being infected ,wear protective glasses if necessary.

7.6 Clean flow cell Both misusing of flow cell and drawing it out frequently all may cause a cut or a permanent damage on surface of the flow cell .Dust on the surface may decline accuracy of measurement .So please don’t draw flow cell out in normal circumstances. Wipe flow cell with lens paper instead of hard material to avoid the cut of surface.Pay special attention to installing direction after cleaning to avoid an unnecessary damage due to an opposite installation .Please see following pictures for your reference:

Attention When wiping flow cell ,do choose the soft material like lens paper to prevent cutting light-hole which will affect the Visible Light Transmission .At the same time ,pay attentions to installing direction.
